Updating to Reggie 3.4
1. mRNA dilution protocol incompatibilities
Before updating to Reggie 3.4 make sure that there are no mRNA plates waiting for dilution.
Ticket #764 adds support for NeoPrep and manual TruSeq. Since this uses less RNA than the dUTP protocol dilution protocol must support "large mixes" (to avoid going below the 1µl limit). This makes the dilution calculations for the mRNA dilution protocol more complex and also incompatible with mRNA plates created be earlier Reggie versions.
2. Create at least one Normalization protocol
The 'RNA concentration normalization' and 'Library preparation design' wizards need at least one protocol with subtype 'Normalization'. The protocol must specify values for the 'RNATargetVolume' (µl) and 'RNATargetAmount' (µg) annotations. It is recommended that normalization protocols are created for the procedures that are going to be used in the lab in the future. For example:
- Manual TruSeq, 50µl, 0.5µg
- NeoPrep TruSeq, 45µl, 0.1µg
